Protagonist Therapeutics (Nasdaq:PTGX) reported third-quarter 2026 financial results alongside updates on its partnered medicines, wholly owned pipeline and share repurchase programme. The company recorded $274.4 million in license and collaboration revenue and net income of $220.0 million for the quarter ended September 30, 2026.
#MIMRYLO approval triggers Takeda payments
Takeda received U.S. Food and Drug Administration approval for MIMRYLO (rusfertide) on August 28 for the treatment of erythrocytosis in adults with polycythemia vera. The medicine was also approved in the United Arab Emirates on September 30, 2026.
Protagonist said the U.S. approval triggered $275 million in payments, comprising a $200 million opt-out payment and a separate $75 million approval milestone. The payments were received in cash in October 2026. Protagonist recognised $266.6 million proportionally in third-quarter revenue in connection with the $275.0 million earned from Takeda.
Following Protagonist’s April 2026 opt-out election, Takeda has exclusive worldwide development and commercialisation rights to MIMRYLO. Protagonist remains eligible for tiered royalties ranging from 14% to 29% on worldwide net sales and up to an additional $875 million in potential future milestone payments. The company said the royalty structure corresponds to an approximately 21% weighted-average royalty rate at $1.5 billion in annual net sales.
#ICOTYDE receives further approvals
During the quarter, ICOTYDE received regulatory approvals in China, Japan and the European Union for psoriasis, following U.S. FDA approval in March 2026. The third quarter was the medicine’s second full quarter of U.S. commercial sales under Protagonist’s collaboration with Johnson & Johnson.
Under that agreement, Protagonist is eligible for tiered royalties ranging from 6% to 10% on global net sales. The company also remains eligible for up to an additional $580 million in potential future milestone payments. Protagonist said a 10% royalty tier applies to incremental annual sales above $4 billion, while the approximate weighted-average royalty rate is 7.25% at $4 billion in annual net sales.
Johnson & Johnson presented two-year Phase 3 results from the ICONIC-TOTAL and ICONIC-LEAD studies in September and October 2026. Protagonist said the results showed responses continuing through Week 112 and a favourable safety profile in adults and adolescents with plaque psoriasis.
#Clinical pipeline plans
Protagonist said it had completed the Phase 1 study of PN-881, an oral IL-17 antagonist peptide for immune-mediated disease. The company reported 24-hour trough concentrations several-fold above its prospectively targeted IC 90 values for IL-17AA, IL-17FF and IL-17AA+FF with once-daily oral dosing. PN-881 was generally well tolerated, with no serious adverse events reported.
Protagonist plans to initiate a comprehensive Phase 2b dose range-finding study in patients with plaque psoriasis in early Q1 2027. The company said Phase 1 data are expected to be presented at a future medical meeting.
The company has prioritised development of orally administered PN-477o, a triple GLP-1/GIP/GCG agonist peptide for obesity, after findings from the subcutaneously administered PN-477sc Phase 1 SAD study. Protagonist anticipates initiating a Phase 1 study of PN-477o in the first half of 2027.
For PN-458o, a dual GLP-1/GIP agonist peptide, IND-enabling studies are ongoing and Phase 1 initiation is anticipated in the second half of 2027. IND-enabling work is also continuing for PN-8047, an oral small molecule hepcidin functional mimetic for polycythemia vera, with Phase 1 initiation anticipated in Q1 2027. Protagonist also said it was advancing discovery programmes involving an oral IL-4Rα antagonist and amylin-based mono- and poly-agonists.
#Financial results and cash position
Cash, cash equivalents and marketable securities stood at $834.6 million as of September 30, 2026, compared with $646.0 million as of December 31, 2025. The company reported $274.4 million in license and collaboration revenue for the third quarter, including $7.8 million from ongoing development services and other revenues in addition to the Takeda-related amount.
For the nine months ended September 30, 2026, license and collaboration revenue was $544.3 million. The figure included $460.5 million in total proportional revenue recognised from Takeda, $33.8 million from ongoing development services, a $50.0 million milestone from Johnson & Johnson following ICOTYDE’s U.S. FDA approval, and other revenues.
Third-quarter net income was $220.0 million, or $3.32 per basic share and $3.07 per diluted share. This compared with a net loss of $39.3 million, or $0.62 per basic and diluted share, in the third quarter of 2025. Net income for the nine months ended September 30, 2026, was $386.6 million, compared with a net loss of $85.8 million for the corresponding period of 2025.
Protagonist said research and development expenses are expected to increase for the remainder of 2026 compared with prior quarters. The company attributed the expected increase to the advancement of PN-881, clinical manufacturing and CMC activities, readiness spending for PN-477o, PN-458 and PN-8047, additional discovery programmes, and higher headcount and stock-based compensation expenses.
#Share repurchase programme
On September 8, Protagonist announced that its Board of Directors had authorised a programme allowing the company to repurchase up to $300 million of its outstanding common stock.
The company intends to fund any repurchases from cash, cash equivalents and marketable securities, as well as royalties and other payments received under its Takeda and Johnson & Johnson collaboration arrangements. The timing and amount of repurchases will depend on market conditions, alternative capital deployment opportunities and other factors.
